He ruled from 705 BC until he died in 681 BC. His name in the Akkadian language means "Sîn has replaced the brothers." This name likely came because his older brothers had died before he was born. He was the son of King Sargon II. Sennacherib spent much of his youth in the royal palace at Nimrud.

Sennacherib helped manage the distribution of gifts and tribute. He sent many letters to his father to report his work. These letters show that he respected Sargon II very much. He always tried to please his father during his time as prince.

He had a hard time controlling the land of Babylonia to the south. A tribal chief named Marduk-apla-iddina II caused many problems for him. This leader retook Babylon and worked with the Elamites. Later, the Babylonians and Elamites killed Sennacherib's eldest son, Aššur-nādin-šumi. Because of this, Sennacherib fought in both regions to win control. In 689 BC, he destroyed the city of Babylon. He also fought a war in the Levant in 701 BC. During this time, he led an invasion of the Kingdom of Judah.

Sennacherib had originally chosen his son Arda-Mulissu to be his heir. However, he changed his mind in 684 BC. He chose his younger son, Esarhaddon, to be the next king instead. Arda-Mulissu was very upset and asked to be the heir again. Sennacherib did not listen to his son's many appeals. In 681 BC, Arda-Mulissu and his brother Nabu-shar-usur killed Sennacherib. They did this because they wanted to take power for themselves. After this, Esarhaddon became the new king of Assyria.
