Barrientos was born on May 30, 1919, in Tarata. His father came from Spain and his mother was Quechua. After his father died, he lived in a Franciscan orphanage. He left the orphanage at age 12 to attend high school. He worked odd jobs to pay for his classes. Eventually, he joined the military academy in La Paz and graduated in 1943. He even earned his pilot's license in 1945.

Barrientos became very famous for his bravery as an Air Force commander. In 1960, a parachute jump for soldiers went very wrong. Three people died when their equipment failed. To show the equipment was not the problem, Barrientos jumped from a plane himself. He used one of the parachutes that had failed before. This brave act made him very popular with many people. He also helped the Revolutionary Nationalist Movement party. He even flew to bring back their leader, Víctor Paz Estenssoro, after a revolution in 1952.

His rise to power happened through a series of big changes. In 1964, he was the vice president under Paz Estenssoro. However, a coup d'état happened just three months later. A coup is when the military takes over the government. Barrientos and Alfredo Ovando became co-presidents of a military junta. Later, Barrientos ran in the 1966 elections. He won and was sworn in as president in August 1966. He was a charismatic leader who spoke Quechua fluently. This helped him connect with many Bolivian farmers.

His time as president included many difficult and serious events. In 1967, a guerrilla group led by Che Guevara arrived in the jungle. Barrientos used the army to fight this group. He also sent troops to deal with miners in the town of Siglo XX. This led to the San Juan massacre on June 24, 1967. During this event, soldiers killed about 30 men and women. In 1968, a scandal happened when his friend Antonio Arguedas disappeared. Arguedas had taken a diary belonging to Che Guevara. This event made people doubt the president's judgment.

Even with these troubles, Barrientos tried to stay close to the people. He traveled to very small and remote villages to talk to them. He wanted to fix his relationship with the workers. Sadly, his life ended very suddenly in 1969. On April 27, his helicopter hit telephone cables near Arque. This crash killed Barrientos, his pilot, and his aide. Some people believe he was assassinated, but no one has proven it. His death marked the end of a very intense era in Bolivia.
