He was born on April 13, 1820, in the town of Tarata. His father was a Spaniard named José Linares. His mother was a woman named Ignacia Melgarejo. His father did not recognize him as a son. Because of this, Melgarejo used his mother's last name. He grew up in a very humble home.

Melgarejo had a very fast rise in the military. He joined the army in 1836 when he was only 16. He started as a simple private. He fought in the War of the Confederation. In 1841, he fought in the Battle of Ingavi.

He showed great bravery during that big battle. This led to his promotion to second lieutenant. He rose through many ranks over the next years. He became a captain in 1850 and a major in 1855. By 1862, he reached the rank of general. This was the highest rank in the Bolivian army.

His path to the presidency was full of many changes. In 1854, he tried to lead a revolt in Santa Cruz de la Sierra. The government crushed his revolt and sentenced him to death. However, some women from high society asked the president for mercy. President Manuel Isidoro Belzu decided to pardon him. Melgarejo then continued his military career for many more years. He helped different presidents take power through various coups. In 1857, he helped overthrow President Jorge Córdova. In 1862, he helped President José María de Achá take control.

Melgarejo became the fifteenth president of Bolivia in late 1864.

He took power after a coup against President Achá. His rule is often called the Sexenio. It lasted for about six years. His time as leader was very difficult for the country. He was known for poor government work. He also treated the indigenous people very unfairly. He signed border treaties with Chile and Brazil in 1866 and 1867. These treaties were very bad for Bolivia in the future. In 1865, he also killed former president Manuel Isidoro Belzu.

His time in power finally came to an end in 1871. People in Bolivia were tired of his harsh actions. General Agustín Morales led an uprising against him in January 1871. A very fierce battle happened in the city of La Paz. The people and the soldiers fought a hard battle. The uprising won and Melgarejo lost his presidency. He fled the country and went to Chile for a few months. Later, he went to Lima, Peru, to find a woman named Juana Sánchez. Sadly, he was shot and killed in Lima on November 23, 1871.
