His father, Pishori Lal Anand, was a prominent lawyer and a freedom fighter. His father was also a scholar who studied various world religions. Dev studied English Literature at Government College, Lahore, before moving to Bombay. In the early 1940s, he held humble jobs to support himself. He worked in a military censor's office for 65 rupees a month. Later, he served as a clerk in an accounting firm for 85 rupees.

His entry into the film industry began with a lead role in 1946. He starred in the film Hum Ek Hain, which focused on Hindu-Muslim unity. Anand's breakthrough came through the actor Ashok Kumar. Anand was a pioneer in film production and genre development. In 1949, he co-founded Navketan Films with his brother, Chetan Anand. The name Navketan means "New Banner." He also formed a famous professional pact with the director Guru Dutt. They agreed to support each other's success in the industry. Together, they created the 1951 crime thriller Baazi. This film is considered the forerunner of the "Bombay Noir" style. This style influenced many urban crime films throughout the 1950s.

Throughout the 1950s, Anand developed a very recognizable acting style. He used a rapid-fire way of delivering dialogue. He also had a unique habit of nodding while he spoke. These mannerisms were widely imitated by many fans. He starred in various genres, including light comedies and social dramas. He also explored darker roles, such as a smuggler in the film Jaal. His acting in the 1958 film Kala Pani earned him his first Filmfare Award for Best Actor. He often worked with famous actresses like Madhubala and Waheeda Rehman.

In the 1960s, Anand transitioned into a celebrated romantic hero. He starred in many popular films, such as Guide and Tere Ghar Ke Samne. He was known for his ability to play complex roles. For example, in the 1961 film Hum Dono, he performed a double role. He played both a young lover and a soldier named Major Varma. His films during this era often featured high production values and memorable music. He continued to find success even as new generations of stars arrived in the late 1970s.

Anand's contributions to culture earned him the highest honors in India. He received four Filmfare Awards, including two for Best Actor. In 2001, the Government of India awarded him the Padma Bhushan. This is the third-highest civilian honor in the country. In 2002, he was presented with the Dadasaheb Phalke Award.
