She attended an open casting call held by John Fearnley. Fearnley served as a casting director for the legendary duo Rodgers and Hammerstein. He was so impressed by her that he ran across the street to find Richard Rodgers. Rodgers then contacted Oscar Hammerstein at home to discuss her potential. This led to Jones becoming the first and only singer under personal contract with the songwriters. She began her Broadway career with a minor role in the musical South Pacific.

Her early stage work allowed her to develop significant professional skills. She appeared in the Broadway production Me and Juliet. In this show, she started as a chorus girl before becoming an understudy for the lead role. Her performances in Chicago earned her high praise from critics. This momentum led to her being cast as the female lead in the 1955 film adaptation of Oklahoma!. Following this success, she starred in several other major musical films. These included Carousel in 1956, April Love in 1957, and The Music Man in 1962.

Jones proved her dramatic depth through a highly acclaimed film performance. In the 1960 film Elmer Gantry, she played a character who was corrupted by the title role. This role was a departure from her usual wholesome image. Director Richard Brooks had initially resisted her casting in the film. However, after seeing her first scene, he predicted she would win an Academy Award. She did indeed win the Academy Award for Best Supporting Actress for this role. This achievement solidified her reputation as a serious dramatic actress.

In 1970, Jones achieved massive television success with The Partridge Family.

This ABC musical sitcom was loosely based on the real-life musical family, The Cowsills. Jones played Shirley Partridge, a widowed mother of five children. The family traveled together in a signature painted bus. The series became a global hit, reaching audiences in over 70 countries. During the show's run, Jones and her co-stars became major pop culture icons. Her stepson, David Cassidy, played her eldest son, Keith, and became a teen idol.

This television era also brought significant musical milestones. Jones and Cassidy performed together on several albums and singles. Their song "I Think I Love You" reached number one on the Billboard Hot 100 chart. This achievement made Jones the first woman to win an acting Oscar and also reach number one on that music chart. She was the second person ever to do so, following Frank Sinatra. The Partridge Family also earned a NARM award for the best-selling single of the year in 1970. Additionally, the group received a Grammy nomination for Best New Artist in 1971.

David Cassidy was Jack's son from a previous marriage, making Shirley his stepmother. Despite the complexities of their family dynamic, they became very close friends. Their bond deepened following Jack Cassidy's death in 1976. Later, Jones married comedian Marty Ingels in 1977, and they remained married until his death in 2015.
