Her career spanned over four decades of work. She performed on theater stages, in motion pictures, and on television. Van Fleet was widely recognized for her ability to portray characters much older than her actual age. This skill required immense discipline and specialized preparation. She often played intense or tragic roles that demanded great emotional depth.

Acting requires rigorous training to master the craft. Van Fleet began her academic journey at the University of California, Berkeley, graduating in 1936. She later pursued a graduate program at the College of the Pacific in Stockton, California. To refine her technique, she moved to New York City. There, she studied with Sanford Meisner at the Neighborhood Playhouse. She also studied with influential figures like Elia Kazan and Lee Strasberg at the Actors Studio. These methods helped her develop the skills used in her professional career.

Van Fleet's professional journey began on the stage in 1944. Her first notable role was Miss Phipps in the production of Uncle Harry in Washington, D.C. She soon moved to Broadway in New York. She performed in several Shakespearean plays, including The Winter's Tale and King Lear. In 1954, she reached a major milestone. She won the Tony Award for Best Featured Actress in a Play. This honor was for her performance as Jessie Mae Watts in the production The Trip to Bountiful.

Her transition to film brought even greater fame. In 1955, director Elia Kazan cast her in her film debut. This movie was a screen adaptation of John Steinbeck's East of Eden. Van Fleet played the character Cathy Ames. Her performance earned her the Academy Award for Best Supporting Actress. She was also nominated for a BAFTA Award for Most Promising Newcomer. This period marked the height of her cinematic success. She appeared in several other films through 1960, such as The Rose Tattoo and Gunfight at the O.K. Corral.

Transforming her appearance was a central part of her work. In the 1960 film Wild River, she played an 89-year-old woman named Ella Garth. Although she was only 44 years old, she underwent a massive transformation. She spent five hours every morning receiving professional make-up application. This process allowed her to convincingly portray a matriarch of the Garth family. Such dedication to character detail defined much of her later work.

Van Fleet's career included many diverse roles across different media. She played the Wicked Stepmother in the 1965 television movie version of Cinderella. She also acted alongside major stars like Paul Newman and Anthony Perkins.

Her life was marked by both professional triumph and personal challenges. She was born in 1915 in Oakland, California. Her father, Roy H. Van Fleet, died in 1919 from a streptococcus throat infection. Her mother, Elizabeth "Bessie" Catherine, worked as a sales lady to support the family. In 1946, Jo married William G. Bales, a professional in the field of modern dance. They remained married until his death in 1990. Van Fleet passed away in 1996 at the age of 80 in New York City.
