Nichols began her professional journey in Chicago, Illinois. She grew up in the suburb of Robbins and attended Englewood High School. From the age of 12, she studied dance at the Chicago Ballet Academy. She started her career as a singer and dancer. She toured the United States and Canada with the bands of Duke Ellington and Lionel Hampton. She also appeared as the principal dancer in the 1959 film version of Porgy and Bess. Her early work included stage roles in plays like Kicks and Co. and Carmen Jones.

Her role on Star Trek became a defining moment in her life. During the first season, Nichols considered leaving the series for a role on Broadway. She even wrote a resignation letter to producer Gene Roddenberry. However, a meeting with Dr. Martin Luther King Jr. changed her decision. King told her that she was a vital role model. He compared her work on the series to the marches of the civil rights movement. Inspired by this, she decided to stay with the show.

Nichols contributed to television history through several significant moments. In the November 22, 1968, episode "Plato's Stepchildren," she shared a kiss with actor William Shatner. This moment is often cited as the first interracial kiss on U.S. television. Although the scene was portrayed as being forced by alien telekinesis, it received almost no dissent. She also expanded her Star Trek legacy through voice work. She provided the voice for Uhura in The Star Trek Animated Series. She later appeared in six Star Trek feature films, including Star Trek VI: The Undiscovered Country in 1991.

Beyond the screen, Nichols was a dedicated advocate for space exploration. From 1977 to 2015, she volunteered her time to promote NASA's programs. She worked to recruit diverse astronauts, specifically focusing on women and ethnic minorities. Her efforts helped bring many pioneers to the space program. These included Dr. Sally Ride, the first American female astronaut, and Colonel Guion Bluford, the first African-American in space. She also helped recruit Dr. Judith Resnik and Dr. Ronald McNair.

Her influence extended to several generations of scientists and performers. Former NASA astronaut Mae Jemison stated that the role of Lieutenant Uhura inspired her to become an astronaut. The famous entertainer Whoopi Goldberg also noted Nichols' influence. Goldberg mentioned that her own character, Guinan, was specially created after she asked for a role on Star Trek: The Next Generation. Nichols' musical career also flourished with two albums. Her 1967 album, Down to Earth, featured standards. Her 1991 album, Out of This World, focused on themes of space exploration.

In her later years, Nichols continued to work across many media formats. She appeared in the television drama Heroes as Nana Dawson in 2007. She also had a recurring role on the soap opera The Young and the Restless. In 2017, she earned a Daytime Emmy nomination for her work on that series. She even voiced characters in popular animated shows like The Simpsons, Futurama, and Gargoyles. Throughout her life, Nichols remained a symbol of possibility for many people.
