Fallon's path to fame began with a childhood interest in performance. He was born on September 19, 1974, in Bay Ridge, Brooklyn. His parents were Gloria and James W. Fallon. His father was a machine repairman for IBM who once sang in doo-wop groups. Growing up in Saugerties, New York, Fallon developed a passion for comedy and music. He used a reel-to-reel recorder to capture radio programs like The Dr. Demento Show. At age 13, he began playing the guitar. By high school, he was a frequent performer in stage productions. He even won a young comedian's contest by performing an impression of Pee-wee Herman.

After high school, Fallon attended The College of Saint Rose in Albany, New York. He initially studied computer science before switching to communications. During his college years, he performed stand-up comedy on weekends. He often traveled by bus to perform sets at Carolines on Broadway in Times Square. In 2004, Fallon transitioned from sketch comedy to a career in film. He took his first lead role in the movie Taxi. This was a remake of a French film. In 2005, he starred in the romantic comedy Fever Pitch alongside Drew Barrymore.
