It is located in the town of Stellenbosch in the Western Cape province. The university sits on the banks of the Eersterivier, also called the First River. It is surrounded by mountains and is located about 50 kilometers from Cape Town. Today, the university is home to approximately 32,000 students. It is organized into 139 different departments. These departments are grouped into 10 specific faculties. These faculties include AgriSciences, Arts and Social Sciences, and Engineering. The university operates across five distinct campuses in the Western Cape. The main campus is located in Stellenbosch. The Bellville Park Campus serves as the Business School. The Saldanha Campus focuses on Military Science. The Tygerberg Campus is dedicated to Medicine and Health Sciences. There is also the Ukwanda Rural Clinical campus for Medicine and Health Sciences. The history of the institution began in 1864. It started as the Stellenbosch Gymnasium and opened on March 1, 1866. In 1870, the first five students matriculated. During the 1870s, the government of the Cape Colony began to prioritize education. This led to more funding and staff for the school. In 1873, four of the original students became the first graduates. They earned a "Second Class Certificate" through distance learning. The institution grew rapidly after 1874 when the government provided subsidies. In 1876, the school formed its first academic senate. By 1878, the first MA degree was completed in South Africa.

As the school grew, its name changed several times. It became Stellenbosch College in 1881. In 1887, it was renamed Victoria College. The path to becoming a full university required a large amount of money. The government required the college to raise £100,000 to gain university status. A wealthy farmer named Jannie Marais provided the necessary funds. He left a bequest before his death in 1915. His gift included conditions regarding language use. He wanted Dutch and Afrikaans to have equal status to English. Because of his support, the institution became Stellenbosch University on April 2, 1918.

The university has a diverse student body and a changing language policy. Historically, it was a predominantly Afrikaans-medium university. However, the demand for English instruction has increased as the student body has grown. In 2021, the university reported that 48.7% of students spoke English as their home language. Meanwhile, 37.4% stated Afrikaans was their home language. The university now promotes multilingualism to ensure equitable access for everyone. This includes using English, Afrikaans, and Xhosa in social and academic contexts. Between 2017 and 2021, the percentage of undergraduates preferring English instruction rose from 68.2% to 80.8%.
