Booth's journey into acting began with a spark of interest in Philadelphia. At age seven, she saw a stage performance that changed her life. She later moved to Connecticut and acted in summer stock plays. As a teenager, she traveled to New York City to find work. She lived at the Rehearsal Club with other young actors. She even used the name Thelma Booth for a while. This was because her father did not want her using the family name for work. She eventually chose the name Shirley Booth for her professional career.

Her career grew steadily through many different types of shows. She made her Broadway debut in the play Hell's Bells in 1925. In that play, she acted alongside Humphrey Bogart. Later, she became very famous for the comedy Three Men on a Horse. This show ran from 1935 to 1937 and brought her great notice. She also worked on radio shows like Duffy's Tavern. She played a funny, clever character on that program. Throughout the 1930s and 1940s, she moved between dramas and musicals.

One of her most important roles was Lola Delaney. She played this character in the drama Come Back, Little Sheba. This role was a huge success on the Broadway stage in 1950. She then played the same role in a movie in 1952. For this movie, she won the Academy Award for Best Actress. She was the first actress to win both a Tony and an Oscar for the same role.

She eventually moved to North Chatham in Cape Cod, Massachusetts. There, she spent her time painting and doing needlework. She lived with her pet poodle and two cats. Even after she stopped acting, her legacy lived on. She has a star on the Hollywood Walk of Fame to honor her work.
