The story follows two children who become close friends. Jim Burden is an orphan from Virginia. He moves to Black Hawk, Nebraska, to live with his grandparents. At the same time, the Shimerda family moves from Bohemia to the same area.

Ántonia Shimerda is the elder daughter in her family. The two children meet and explore the colorful summer land together. They share many adventures, like the time Jim used a shovel to kill a rattlesnake. Jim also helps Ántonia's family by teaching her sisters to read English. These early years leave strong impressions on them for their whole lives.

Life on the frontier was often very hard for these pioneers. The Shimerda family had a very difficult first year. They did not have a real house at first. Instead, they lived in a cave in the earth. Mr. Shimerda missed his old home and his violin in Bohemia. The pressures of the new life were very heavy for him. Eventually, he died, and Ántonia had to work the land with her brother. They eventually built a log cabin to live in.

As the characters grow up, their paths change in many ways. Ántonia works as a housekeeper for families like the Harlings. She even works for a couple named the Cutters. Jim goes to school and becomes the valedictorian of his class. He eventually studies law at Harvard in Boston. He becomes an attorney for a western railroad. Even though they are far apart, Jim and Ántonia stay connected. Years later, Jim visits Ántonia on her farm in Nebraska. He meets her husband, Anton Cuzak, and her many children.

This book changed how people wrote about America. Before this, many stories focused on wealthy families. Cather chose to write about "hired girls" and immigrant families instead. This was a radical and new way to tell a story. The novel is divided into five different sections called "Books." These include "The Shimerdas," "The Hired Girls," "Lena Lingard," "The Pioneer Woman's Story," and "Cuzak's Boys."

The book is still a very important part of American literature today.
