He was the thirteenth child in a very large family of fifteen children. After his father died when Luther was 18, he used his inheritance to buy land. He sold the rights to his first potato for $150. In 1875, he used that money to travel to Santa Rosa, California. There, he set up a greenhouse and experimental fields. He was inspired by the ideas of Charles Darwin to study how plants change.

Many of his creations are still used around the world today.

Stark's company, Stark Bro's Nurseries & Orchards Co., offered Burbank $9,000 to support his research. This partnership helped make Burbank's work profitable. He also received grants from the Carnegie Institution between 1904 and 1909. Even though some academic critics questioned his scientific methods, Andrew Carnegie supported his research on hybridization.

Burbank faced criticism from the scientific community of his time. Many professors felt he was not a scientist in the academic sense. This was because he did not keep the detailed, careful records required by formal research. He focused more on creating useful plants than on studying their biological mechanisms. However, his lack of record-keeping may have been due to the era's difficulties. There was no easy way to legally protect inventions at that time. He also felt his time was better spent in the garden than writing in books.

Burbank passed away on April 11, 1926, due to heart failure. Before his death, he asked his wife, Elizabeth, to ensure his work continued. He suggested that Stark Bro's Nurseries & Orchards Co. was the best organization for his legacy. Elizabeth eventually sold the property and various rights to the Stark company. Much of his experimental farm was later used by the Santa Rosa Junior College. Today, the site is a public park and a free showplace for the city. His many unmarketed experiments, like hybrid roses and different types of plums, remain part of his vast botanical legacy.
