He has built a career by portraying a wide range of characters, from innocent heroes to complex villains. Much of his fame stems from his creative partnership with director David Lynch. This collaboration helped define his screen persona as someone who could navigate strange or mysterious worlds. Through decades of work, MacLachlan has become a recognizable figure in both mainstream media and cult classics.

MacLachlan's path to professional acting began with musical training and local theater. His mother, Catherine, was a public relations director who also led a youth theater program in Yakima, Washington. She introduced him to stage acting when he was 15 years old. Before focusing entirely on drama, he studied piano from age 9 to 14 and also practiced classical singing. He eventually attended the University of Washington, where he studied in the Professional Actor Training Program. In 1982, he graduated cum laude with a BFA, or Bachelor of Fine Arts, in drama.

His professional film career began with very small roles, such as an extra in the 1980 film The Changeling. For this role, he was paid only $10. His first major opportunity arrived in 1984 when he was cast as Paul Atreides in the science fiction film Dune. Although the film received poor reviews and did not succeed commercially, it led to a life-changing meeting. While performing in a play in Seattle, he was recommended to producer Dino De Laurentiis. This led to his meeting with David Lynch, a director who shared a similar Pacific Northwest background.

The partnership between MacLachlan and Lynch became a cornerstone of his career. Lynch cast him as Jeffrey Beaumont in the 1986 film Blue Velvet, which received much better reviews than Dune. This relationship continued through several major projects, including the iconic television series Twin Peaks from 1990 to 1991. In this series, MacLachlan played Special Agent Dale Cooper, a role that earned him a Golden Globe and two Primetime Emmy Award nominations.

Lynch described MacLachlan's ability to play characters who are interested in the mysteries of life.

Throughout the 1990s and 2000s, MacLachlan expanded his repertoire into many different genres. He played Ray Manzarek, a musician in the band The Doors, in the 1991 film of the same name. In 1994, he took on a different kind of role as the antagonist Cliff Vandercave in The Flintstones. He also appeared in the 1995 film Showgirls, which was famously criticized by reviewers but later embraced for its campiness. In the early 2000s, he became a recurring character on the series Sex and the City, playing Dr. Trey MacDougal. He also moved into stage acting, making his Broadway debut in 2003 in the play The Caretaker.

In more recent years, MacLachlan has embraced voice acting and modern television adaptations. He provided the voice for Riley's father in the Pixar animated films Inside Out and Inside Out 2. He also appeared in the television series Portlandia as the Mayor of Portland from 2011 to 2018. Currently, he stars in the series Fallout, which is an adaptation of a popular video game. His work on this series has been noted for allowing him to show both goofy and menacing sides of his acting skill. In 2024, his contributions to the industry were recognized with the Canal+ Icon Award at Canneseries.

MacLachlan's career demonstrates how a single creative connection can launch a lasting legacy. His ability to transition from the intense psychological worlds of David Lynch to family-friendly animation shows immense professional range. He has successfully navigated the shifts from film to television and from live theater to voiceover work. By maintaining a presence in diverse media, he has remained a relevant and respected figure in the global entertainment industry for over forty years.
