Khoo's career moved through many different steps. He started as an apprentice clerk at OCBC in 1933. He worked hard and rose to become a deputy general manager. In 1960, he helped found a new bank called Maybank in Kuala Lumpur. This bank grew very fast and had over 150 branches in just three years. Later, he became a "white knight" for Standard Chartered. A white knight is someone who helps a company during a hard time. He bought a 5% stake to stop Lloyds Bank from taking it over. Eventually, he owned almost 15% of that bank.

Khoo's journey began with his education in Singapore. He attended St Joseph's Institution starting in 1930. He finished his studies up to standard eight. He married at the age of 17. His early work at OCBC led him to serve as chairman of the Central Provident Fund board in 1958. He left OCBC in 1959 after a difference of opinion. From there, he moved into founding banks and buying large properties. He even served as a senator in the Malaysian parliament from 1964 to 1965.

There were many important numbers in his life. In 1963, his bank bought the Goodwood Park Hotel for S$4.8 million. In 1968, he bought several properties for S$50 million. He once used funds from the National Bank of Brunei to buy the Southern Pacific Hotel Corporation. This led to a difficult time when the bank closed after Sultan Hassanal ordered an investigation. Khoo was never charged with a crime during this time. In 2004, Forbes ranked him as the 108th richest person in the world. He died at Mount Elizabeth Hospital from a heart attack.

Khoo Teck Puat is remembered for his many gifts to others. He started the Khoo Foundation in 1981 with S$20 million. This charity helped many people through different projects. He gave S$125 million to help build Alexandra Hospital @ Yishun. In 2007, the hospital was renamed Khoo Teck Puat Hospital to honor him. He also gave S$80 million to the Duke-NUS Medical School. Another S$10 million went to a charity fund for children and the elderly. His money continues to help people stay healthy today.
