He was a key figure in early French attempts to settle Florida. Ribault was a Huguenot, which is a type of Protestant. He worked under a French admiral named Gaspard de Coligny. In 1562, Coligny chose him to lead an expedition to the New World. Ribault left France on February 18 with 150 colonists. They were likely guided to Florida by a Portuguese pilot named Bartolomeu Borges. This first trip helped the French start to explore the coast.

Ribault first explored the mouth of the St. Johns River. He called it the "River May" because he found it in May. He even put up a stone column to claim the land for France. The fleet then sailed north to explore more of the coastline. They eventually reached the Port Royal Sound in South Carolina. Ribault decided to build a settlement on Parris Island. He named this small fort Charlesfort to honor King Charles IX. He left 27 men there under Albert de la Pierria to guard it.

Ribault sailed back to France to get more supplies.

While Ribault was away, things went very wrong at Charlesfort. A fire destroyed most of their food and supplies. There was also fighting among the soldiers. The survivors tried to sail back to France in a small boat. Most of them died before they reached the English coast. Meanwhile, Ribault was busy with wars in France. He even spent time in the Tower of London as a spy. Later, he organized a second voyage to help the French colonies. He planned to build a new settlement called Fort Caroline near Jacksonville.

In 1565, Ribault finally sailed with 800 settlers and five ships. He arrived in Florida on August 28. However, the Spanish were already planning to remove the French. A Spanish leader named Pedro Menéndez de Avilés arrived in early September. A large hurricane struck the coast on September 11. The storm was very powerful and destroyed Ribault's fleet. The ships were driven south to the Canaveral coast. Many sailors survived the storm but were left hungry and lost. They walked 14 miles to reach Matanzas Inlet.

At the inlet, the Spanish soldiers found the Frenchmen. Following orders from King Philip II of Spain, the Spanish killed the French prisoners. Because of this, the place was named Matanzas, which means "slaughters." About 350 of Ribault's men died in these events. Today, we remember Ribault in many ways. A stone column replica stands at the Fort Caroline National Memorial. His name is also on schools and streets in Jacksonville. In 2018, researchers even found his flagship, La Trinité, near Cape Canaveral.
