Cassini’s early career began in Italy at the Panzano Observatory. There, he worked with the amateur astronomer Marquis Cornelio Malvasia. In 1650, the senate of Bologna appointed him as the principal chair of astronomy at the University of Bologna. During this time, he studied under scientists like Giovanni Battista Riccioli and Francesco Maria Grimaldi. He also worked as a hydraulic engineer and an inspector of fortifications for the Pope. These diverse roles helped him master both the math of the stars and the physics of Earth's waters.

One of Cassini's most important early achievements involved the Sun and Earth. He used a camera obscura effect at the San Petronio Basilica in Bologna. He moved a pinhole gnomon high into the church's vaults, 66.8 meters above the floor. This created a large image of the Sun's disk on the meridian line. By measuring the changes in the Sun's apparent size throughout the year, he gathered vital data. He concluded that the Earth moves in an elliptical orbit around the Sun. This supported the heliocentric theory proposed by Johannes Kepler.

In 1669, Cassini moved to France to help establish the Paris Observatory.

He served as the director of this observatory for forty-one years until his death. He also served as an astronomer and astrologer to King Louis XIV. While he studied astrology in his youth, he focused almost entirely on astronomy during his time in Paris. He became so integrated into French life that he was known as Jean-Dominique Cassini. His work at the observatory helped advance the Scientific Revolution through precise observation.

Cassini made several groundbreaking discoveries regarding the planets. He discovered four satellites of Saturn: Iapetus, Rhea, Tethys, and Dione. He also identified a gap in Saturn's rings, now known as the Cassini Division. In 1675, he discovered this division within the ring system. He also shared credit with Robert Hooke for observing the Great Red Spot on Jupiter. Additionally, he was the first to observe differential rotation in Jupiter's atmosphere. These findings provided a much clearer picture of how giant planets function.

His mathematical skills allowed him to measure the scale of the solar system. In 1672, Cassini and his colleague Jean Richer performed simultaneous observations of Mars. They worked from different locations, with Richer in French Guiana. By computing the parallax, they determined the distance to Mars. This allowed scientists to estimate the absolute dimensions of the entire solar system.

He also developed methods to determine longitude by using the eclipses of Jupiter's moons as a celestial clock.

On Earth, Cassini's engineering and mapping changed how nations understood their borders. He used the technique of triangulation to begin a project to create a topographic map of France. This was the first time an entire country was mapped in such detail.

The project was so massive that it was continued by his son, Jacques, and his grandson, César-François. It was eventually published as the Carte de Cassini.
