She is a central figure in Nollywood, the massive Nigerian film industry. Nnaji has appeared in over 200 movies throughout her extensive career. Her work spans acting, directing, and producing films. She is also a dedicated women's activist. She uses her platform to advocate for the rights of Nigerian girls. She specifically speaks against early marriages and the abuse of women. Nnaji describes herself as a feminist. To her, feminism means a woman has the right to make her own choices.

Nnaji was born on May 3, 1979, in Mbaise, Imo State. This location is in the southeast region of Nigeria. She grew up in Lagos, which is in southwest Nigeria. She was the fourth of eight children in her family. Her father worked as an engineer. Her mother was a nursery school teacher.

Nnaji attended Methodist Girls College in Yaba, Lagos. She later went to the University of Lagos. There, she earned a bachelor's degree in creative arts. While she was a student, she began auditioning for acting roles.

Her professional journey began as a child actor. At age eight, she appeared in the television soap opera Ripples. In 1998, at age 19, she entered the film industry with the movie Most Wanted. Since then, she has starred in many notable films. These include Last Party, Mark of the Beast, and Ijele. In 2010, she starred in the film Ijé: The Journey. In 2004, she also explored music. She signed with EKB Records, a Ghanaian record label. She released a debut album titled One Logologo Line. This album featured a mix of R&B, hip-hop, and urban music.

Nnaji has achieved many historic milestones in her career. In 2005, she won the Africa Movie Academy Award (AMAA) for Best Actress in a Leading Role. She was the first actor to ever win this specific award. In 2001, she became the first actor to win Best Actress at the City People's Awards. As a producer and director, Nnaji has pushed Nigerian cinema onto the global stage. In 2015, she produced her first movie, Road to Yesterday. This film won Best Movie Overall - West Africa at the 2016 Africa Magic Viewers' Choice Awards. In 2018, she made her directorial debut with the movie Lionheart. This film was acquired by the streaming service Netflix. It became the first Netflix original film from Nigeria. Lionheart was also the first Nigerian film submitted to the Oscars. It was selected by the Nigerian Oscars Selection Committee for the Best International Feature Film category.

The Oscar submission for Lionheart faced a unique challenge regarding language rules. The Academy rules require eligible movies to have a predominantly non-English dialogue track. Because Lionheart used mostly English, it was disqualified from the race. Nnaji explained that Nigeria has over 500 languages. English serves as the official language to connect such a diverse population. Filmmaker Ava DuVernay questioned this decision on social media. Scholar Kovie Biakolo also noted that Nigeria was penalized for using the language imposed by British colonial history. This debate highlighted issues regarding cultural diversity and inclusivity in global film awards.

Beyond film, Nnaji has been a successful model and entrepreneur. In 2004, she was voted the face of Lux in Nigeria.
