The university is a massive institution with significant resources. In 2024, Duke spent more than $1.5 billion on research. Its endowment is valued at $11.9 billion. This makes it the twelfth-wealthiest private academic institution in the country.

The university's origins date back to 1838.

It began as Brown's Schoolhouse in Randolph County. A group known as the Union Institute Society organized the school. This group consisted of Methodists and Quakers. The school underwent several name changes as it grew. It became Union Institute Academy in 1841. It was later named Normal College in 1851. By 1859, the institution was called Trinity College. In 1892, the college moved to Durham. This move was made possible by the generosity of Julian S. Carr and Washington Duke. Both men were successful in the tobacco and electrical industries.

Significant financial contributions shaped the modern university.

Julian S. Carr donated a 62-acre tract of land called Blackwell Park. This land is now known as the East Campus. Washington Duke also provided substantial funding for the school. He gave $85,000 to help build the institution. He also made several large contributions in 1896, 1899, and 1900. Washington Duke required that women be treated on an equal footing with men. In 1924, his son, James B. Duke, established the Duke Endowment. This was a $40 million trust fund. The fund supported hospitals, orphanages, and several colleges. Following this, the school was renamed Duke University to honor Washington Duke.

The campus is divided into distinct architectural areas.

The West Campus features Collegiate Gothic architecture. This style is characterized by grand, stone buildings. The Duke Chapel sits at the center and highest point of this campus. The East Campus uses a Georgian-style architecture. This area serves as the home for all first-year students. The university also maintains a marine lab in Beaufort.

In the 1920s, the university had policies that excluded Black students. These policies restricted access to certain campus facilities. Students began to petition for change in 1948. The undergraduate college officially desegregated in 1963. The university enrolled its first Black graduate students in 1961. The teaching staff did not become integrated until 1966. In 1972, the separate Woman's College merged with Trinity College. This merger allowed men and women to study together in one liberal arts college. The university has since worked to recruit minority faculty and students.

Athletics are a prominent part of the Duke experience.

In 2006, three lacrosse players were falsely accused of a crime. A local official later declared them innocent. The university administration was criticized for its actions during this time. In 2015, the Office of Research Integrity found research misconduct by a former professor. The researcher had falsified data in cancer studies. Additionally, the university has dealt with the removal of controversial monuments. In 2017, a statue of Robert E. Lee was removed from the Duke Chapel entrance. These events show how the university continues to navigate its role in society.
