The Treaty of Tordesillas in 1494 divided the world into spheres of influence for Spain and Portugal. This linked missionary work to colonial policy. In the Netherlands, the Catholic Church was suppressed from 1592 to 1853. During this time, congregations were called "stations" or "staties."

Protestantism introduced new missionary patterns after the Reformation. Early Protestant churches initially focused on spreading their faith within Christian lands. Eventually, they began sending missionaries to previously unreached people. In North America, Jonathan Edwards became a missionary to the Housatonic Native Americans in 1751. He advocated for them against cultural imperialism. In the 19th century, the American Board of Commissioners for Foreign Missions was chartered in 1812. The Moravian Church also began sending missionaries in 1732. In the Methodist tradition, Thomas Coke is known as the "Father of Methodist Missions." He worked to increase support for missionary workers.

The Baptist movement also developed strong global missionary organizations. In 1792, twelve ministers founded the Baptist Missionary Society in England. William Carey became its first missionary. He traveled to Calcutta in 1793. Carey used geographic and ethnographic data to map unreached populations. His work is often called the foundation of modern missions. Throughout history, missionaries have interacted deeply with local cultures. Some learned indigenous languages like Quechua, Guarani, and Nahuatl.
