Pyridine is a basic heterocyclic organic compound with the chemical formula C5H5N.

It is a colorless liquid that often appears yellow if it is impure or old. Pyridine has a very distinctive and unpleasant fish-like smell. It is also highly flammable and acts as a weakly alkaline substance. This compound is incredibly important in modern chemistry. It serves as a building block for many commercial products. These include essential vitamins, various pharmaceuticals, and many agrochemicals used in farming.

To understand its chemistry, one must look at its molecular structure. Pyridine is structurally related to benzene, which is a famous six-sided carbon ring. In pyridine, one methine group in the benzene ring is replaced by a nitrogen atom.

This creates a planar, six-sided ring that follows the Hückel criteria for aromatic systems. However, the presence of the nitrogen atom changes how electrons are distributed. The nitrogen creates a negative inductive effect. This causes the electron density to be uneven across the ring. Because of this unevenness, pyridine has a dipole moment. It also has a lower resonance energy than benzene. Benzene has a resonance energy of 150 kJ/mol, while pyridine has only 117 kJ/mol.

The nitrogen atom in the ring plays a very specific role in how the molecule reacts. The ring atoms are sp2-hybridized. The nitrogen atom uses its unhybridized p orbital to participate in the π-bonding aromatic system. Crucially, the nitrogen also possesses a lone pair of electrons. This lone pair sits in an sp2 orbital that projects outward from the ring. This lone pair does not contribute to the aromatic system itself. Instead, it makes the nitrogen atom ready for electrophilic attack. This allows pyridine to support the formation of new chemical bonds quite easily.

The history of pyridine began with early observations by alchemists. They likely produced it by heating animal bones and organic matter. The first formal scientific documentation came from the Scottish scientist Thomas Anderson in 1849.

Anderson was examining oil produced by heating animal bones at high temperatures. He isolated a colorless liquid with a bad odor. Because the substance was so flammable, he named it pyridine. The name comes from the Greek word "pyr," meaning fire. Later, scientists Wilhelm Körner and James Dewar suggested its structure in the late 1800s. They proposed that it was derived from benzene by replacing a C–H unit with nitrogen. This idea was later confirmed through experiments involving the reduction of pyridine to piperidine.

Methods for making pyridine have evolved significantly over time. Historically, it was extracted from coal tar. This was a very inefficient process because coal tar contains only about 0.1% pyridine. This required many stages of purification to get a usable amount. A major breakthrough occurred in 1924 when Aleksei Chichibabin invented a new synthesis. His method used inexpensive reagents and remains a foundation for industrial production today.

One common industrial route involves the Chichibabin synthesis. In this process, formaldehyde and acetaldehyde are used to form acrolein. The acrolein then condenses with more acetaldehyde and ammonia to create dihydropyridine. Finally, this is oxidized to produce pyridine. This reaction typically happens in a gas phase at temperatures between 400 and 450 °C.

Today, the scale of pyridine production is massive. As of 2016, approximately 20,000 tons of pyridine are synthesized worldwide every year. In 1989, the global production was recorded at 26,000 tonnes. Production has grown significantly in recent years, with mainland China reaching a capacity of 30,000 tonnes in the early 2000s. Many large companies produce this chemical, including Evonik Industries and Imperial Chemical Industries. The joint venture Vertellus is currently recognized as the world leader in production. Many other derivatives, such as methylpyridines, are also produced on a very large scale.

Even though it is a laboratory chemical, pyridine appears in trace amounts in the world around us. It is found in the volatile organic compounds produced during roasting and canning. You might find tiny amounts in roasted coffee, fried chicken, potato chips, or fried bacon. It has also been detected in sunflower honey and Beaufort cheese. In the environment, it can be released by industrial processes like steel manufacture or coal gasification. For example, groundwater near a coal gasification plant once showed levels of 53 μg/m3. In the United States, about 43,000 people work in jobs where they come into contact with pyridine.
