Organic synthesis

Organic synthesis is a branch of chemical synthesis focused on building organic compounds. These compounds are molecules composed of hydrogen, carbon, oxygen, and nitrogen atoms. These atoms are held together by covalent bonds. This field is essential for many scientific areas. It is vital for the medical and pharmaceutical industries. Scientists use these methods to create medicines on an industrial scale.

One major goal is total synthesis. This is the complete construction of a molecule from simple, natural precursors. There are two main ways to do this. In a linear synthesis, a chemist performs several steps in a sequence. Each step creates a new compound called a synthetic intermediate. For more complex molecules, a convergent approach is often better. In this method, scientists prepare several key intermediates separately. They then combine these pieces to form the final product.

Chemists also use stereoselective synthesis to manage molecular shape. Many natural products are chiral, meaning they have specific spatial arrangements. These molecules often exist as enantiomers, which are mirror-image versions of each other. Some syntheses produce a racemic mixture, which contains both enantiomers. Stereoselective synthesis allows chemists to direct a reaction to produce only one specific enantiomer. This provides a more diverse choice of pure materials. Notable advancements include stereoselective hydrogenations and asymmetric epoxidation.

Planning a synthesis often requires a retrosynthetic framework. Developed by Elias James Corey, this method involves planning a molecule backward. A chemist starts with the final product and works toward simpler components. They use graphical schemes with retrosynthetic arrows to visualize the design. This process breaks the parent structure into achievable parts. This systematic approach helps ensure the design follows standard chemical rules. Corey received the Nobel Prize in 1990 for this work.

During the process, scientists must use specific tools to manage reactions. Many reactions require heat to increase their speed. To prevent the solvent from boiling away, chemists use a reflux condenser. This is a piece of glassware with water inlets and outlets. The flowing water cools escaping vapors so they condense back into the flask. This ensures the product is contained during reflux or recrystallization.

Separation and purification are also critical steps in the lab. If the product is a liquid, chemists use liquid-liquid extraction. This technique relies on the principle that "like dissolves like." Non-polar compounds dissolve in non-polar solvents. Polar compounds dissolve in polar solvents. By using immiscible solvents, chemists can separate the product based on density and polarity.

If the product is a solid, scientists use filtration. Gravity filtration uses a funnel and paper to let liquid pass through. Vacuum filtration is faster and uses suction to pull liquid through a Büchner funnel. This leaves the solid product on the filter paper.

Modern science is moving toward automated organic synthesis. Researchers use software to create processes that require no human involvement. This includes flow chemistry, where substrates are continually fed into a reaction. This can increase the total yield of the product. An example is the Autosyn system developed by SRI International. This machine can synthesize many FDA-approved small molecule drugs.

Finally, chemists must perform characterization to verify their results. Characterization is the measurement of a compound's physical and chemical properties. This ensures the structure is correct. Common methods include nuclear magnetic resonance (NMR) and mass spectrometry. They also use Fourier-transform infrared spectroscopy (FTIR) and melting point analysis. These tools provide the structural information needed to confirm a successful synthesis.
