Non-nucleophilic base

physical science Maturity 11-13 Next article →

Chemists often need to move tiny parts of a molecule. These parts are called protons. A base is a tool that can take a proton away. Most bases are also nucleophiles. A nucleophile is a part that likes to grab onto other things. Sometimes, a chemist only wants to take a proton. They do not want the base to grab other parts. This is why they use non-nucleophilic bases.

These special bases work because they are bulky. Being bulky means they take up a lot of space. Imagine a person trying to reach for a tiny coin. If they wear huge, puffy gloves, they can still touch the coin. But the gloves stop them from grabbing a large ball. These bases are the same way. They are big enough to stop other reactions. They can reach a small proton, but they cannot grab larger parts.

Scientists have found many kinds of these tools. Some bases are of moderate strength. For example, N,N-Diisopropylethylamine is a base called Hünig's Base. It has a pKa of 10.75. Another one is 1,8-Diazabicycloundec-7-ene, or DBU. DBU has a pKa of 13.5 and helps with E2 elimination reactions. There is also 1,5-Diazabicyclo(4.3.0)non-5-ene, which is similar to DBU. Some are even weaker, like 2,6-di-tert-butylpyridine with a pKa of 3.58.

There are also very strong non-nucleophilic bases. These are usually anions, which are particles with a charge. Lithium diisopropylamide, or LDA, is a very strong base. It has a pKa of 36. LDA is often used to make something called an enolate. This happens during a Claisen ester condensation. Other strong bases include sodium hydride and potassium hydride. These are dense, salt-like materials. They do not dissolve in liquid and work by surface reactions.

Other tools include silicon-based amides. These include NaHMDS and KHMDS. There is also lithium tetramethylpiperidide, which is called a harpoon base. Some bases are in the middle. Sodium tert-butoxide and potassium tert-butoxide have a pKa around 17. They are strong, but they still have a little bit of nucleophilicity. This means they can still grab things a small amount. Each tool helps a chemist do a specific job in a lab.
