Born Maria Luisa Ceciarelli in Rome on November 3, 1931, she adopted her professional name from her mother's maiden name, Vittiglia. Vitti was known for her unique ability to portray complex emotions on screen. She was often described as having a "stunning" screen presence. Upon her death, the Italian culture minister, Dario Franceschini, honored her by calling her "the Queen of Italian cinema." Her career spanned several decades and included work in film, television, and theater.

Her journey into professional acting began with rigorous training and small roles. After performing in amateur productions as a teenager, she attended Rome's National Academy of Dramatic Arts. She graduated from the academy in 1953. Her early career involved a variety of tasks, including voice work. For example, she dubbed the voice of Rossana Rory in the 1958 film Big Deal on Madonna Street. She also worked with an Italian acting troupe that toured Germany. Her first major stage appearance in Rome was in a production of Niccolò Machiavelli's La Mandragola.

Vitti's career reached new heights through her collaboration with director Michelangelo Antonioni. They worked together on several films that explored themes of alienation, which is a feeling of being isolated from others. In the 1960 film L'Avventura, she played a detached protagonist. This role helped make her an international star. She also starred in Antonioni's La Notte in 1961 and L'Eclisse in 1962. Later, she appeared in his 1964 film Red Desert, playing the character Giuliana. Antonioni noted that Vitti inspired him, even though the roles he gave her were often very different from her own personality.

As her fame grew, Vitti worked with many famous international actors. She appeared alongside Marcello Mastroianni, Alain Delon, and Richard Harris. She also acted in English-language films like the spy spoof Modesty Blaise in 1966. This film featured Terence Stamp and Dirk Bogarde. Interestingly, Vitti often resisted starring in American films. A New York Times report noted she disliked long travel, especially by air. She also felt her English was not of a high enough standard. This preference kept much of her most important work centered in Europe.

In the 1970s, Vitti found great success in different genres, including romantic comedies. She starred with Marcello Mastroianni in the successful film Dramma della gelosia, also known as The Pizza Triangle, in 1970. During this decade, she also worked with her partner, cinematographer Carlo Di Palma. She appeared in several films he directed, such as Teresa the Thief in 1973. Her versatility allowed her to move between intense dramas and lighter comedies. This ability to adapt helped her maintain a long and steady career in a competitive industry.

Throughout her life, Vitti received numerous prestigious awards for her talent. She won five David di Donatello Awards for Best Actress. She also earned seven Italian Golden Globes for Best Actress. Her excellence was recognized internationally with a Career Golden Globe and a Venice Film Festival Career Golden Lion Award. In 1984, France honored her with the Order of Arts and Letters. This was a significant recognition of her cultural impact. She was later raised to the rank of Commander of that same order in 1995.

In her later years, Vitti explored new creative paths behind the camera. In 1989, she attempted writing and directing with the film Scandalo Segreto in 1990. She also returned to her roots in the theater as an actress and a teacher. Vitti lived in Rome for much of her life. She passed away on February 2, 2022, at the age of 90. Her death was due to complications from Dementia with Lewy bodies disease. She remains a central figure in the history of Italian film and art.
