Born Alphonsine Rose Plessis on January 15, 1824, she grew up in Normandy, France. Her parents were Marin Plessis and Marie Plessis, who was born Marie Deshayes. At age 15, she moved to the city of Paris to work. She initially found employment in a dress shop. As she grew into her late teens, she realized that wealthy men would pay for her company. This led her to enter the profession of a courtesan. To sound more like a member of the nobility, she added the name "Du" to her surname.

Duplessis did not just rely on her physical appearance to succeed. She was known as an intelligent and witty woman. She took great care to learn how to read and write. She also studied current world events to stay informed. This education allowed her to participate in complex conversations. She could hold her own when speaking with politicians, artists, and writers. This skill helped her maintain her status in the high-society circles of Paris.

Her social life was very active and varied. She hosted salons, which are gatherings where people meet to socialize and discuss ideas. These events brought together many influential people from different fields. Duplessis also enjoyed attending the opera and riding in the Bois de Boulogne. She was the mistress of several famous men during her life. This included the writer Alexandre Dumas fils from 1844 to 1845. She was also believed to be the mistress of the composer Franz Liszt.

In the summer of 1844, she met a significant patron named Count Gustav von Stackelberg. He was a 78-year-old man who was very wealthy. He received a pension from the Tsar and they met at a spa resort in Bagnères-de-Luchon. Because of their relationship, he provided her with a magnificent six-room apartment. This apartment was located on the Boulevard de la Madeleine in Paris. Duplessis was also briefly married to a French nobleman and army officer named Count Édouard de Perregaux.

Despite her many connections, her life ended quite young. Duplessis died of tuberculosis on February 3, 1847. She was only 23 years old at the time of her death. Her husband, the Comte de Perregaux, and Count Stackelberg were both present. Her funeral was held at the Montmartre Cemetery in Paris. It was a large event attended by hundreds of people. However, her death also left financial troubles. Within a few weeks, her personal belongings were auctioned off to pay her debts.

Her legacy grew significantly after she passed away. Alexandre Dumas fils wrote a romantic novel in 1848 titled *La Dame aux Camélias*. The main character, Marguerite Gautier, was based on Duplessis. In the book, Dumas used himself as the character Armand Duval. This story was later adapted into a play in 1852. This play inspired the famous 1853 opera *La traviata* by the composer Giuseppe Verdi. Many films have since used her life as inspiration. A journalist named Romain Vienne later wrote a book to clarify the true facts of her life against many legends.
