He was born in 1930 to a peasant family. His family grew coffee. During a time of fighting in Colombia, Manuel joined a group of fighters. He took the name Manuel Marulanda to honor a dead leader. His group lived in a place called Marquetalia.

In 1964, the Colombian Army attacked Marquetalia. This was called Operation Marquetalia. The attack did not stop the fighters. Instead, they moved and formed a new group. In 1966, they became the FARC. Manuel and Jacobo Arenas led this group.

People called Manuel "Tirofijo." This was a nickname from his comrades. The United States put a $5 million reward on him. They wanted him caught. They said he was a terrorist and a kidnapper. He was never caught by the law. Manuel died of a heart attack on March 26, 2008. Alfonso Cano became the new leader after him.
