Glutathione

Glutathione is a very important molecule found in many living things.

It is an antioxidant found in plants, animals, and fungi. It even exists in some bacteria and archaea. This molecule is a tripeptide, which means it is made of three amino acids. These three parts are glutamate, cysteine, and glycine. It helps protect cells from many types of damage. It can stop harm caused by heavy metals and peroxides. It also fights reactive oxygen species and free radicals.

Making glutathione is a careful two-step process. First, the cell must make a piece called gamma-glutamylcysteine. This step uses L-glutamate and L-cysteine. An enzyme called glutamate-cysteine ligase helps this happen. This is the rate-limiting step, which means it controls how fast the whole process goes. Next, the cell adds glycine to that piece. An enzyme called glutathione synthetase handles this second step. This process uses energy called adenosine triphosphate to work.

Most animal cells can make their own glutathione. However, the liver is the most essential place for this work. Scientists found this by looking at special mice. These mice were made without the ability to make glutathione in their livers. These mice died within one month of birth. This shows how much the body needs this molecule. In humans, we make it too, but some other living things do not. For example, some members of the Fabaceae family do not make it.

Glutathione is very common inside animal cells. It is the most abundant non-protein thiol compound in these cells. The amount inside a cell can range from 0.5 to 10 mmol/L. It is much more concentrated inside the cell than outside. In fact, levels inside can be 1000 times higher than in fluids outside the cell. Most of the glutathione is in a reduced state called GSH. Only a small part is in the oxidized state called GSSG.

Scientists are finding new ways to use this knowledge. Some types of cancer cells have much higher levels of glutathione. Lung, larynx, mouth, and breast cancers can have levels between 10 and 40 mM. Because of this, researchers are making tiny drug delivery systems. These are often called micro-nanogels. These gels use disulfide bonds to hold medicine inside. When the gels enter a cancer cell, the high glutathione causes a thiol-disulfide exchange reaction. This breaks the bonds and releases the medicine exactly where it is needed.
