Diego was born in Porto Santo, Portugal, around 1479 or 1480. He spent his whole life working for the kings of Castile and Aragón. His main goal was to win back the special titles his father once held. These titles were taken away in the year 1500.

Diego worked hard to reach his goals. His marriage helped him a great deal. He married María de Toledo y Rojas. She was the niece of the 2nd Duke of Alba. She was also a cousin to King Ferdinand. This connection to the royal family gave Diego more power.

He spent many years trying to prove his family deserved their rights. He was a page at the Spanish court in 1492.

In August 1508, Diego was named Governor of the Indies. He arrived in Santo Domingo in July 1509. He built a large home there called the Alcázar de Colón.

This grand building still stands today in the Dominican Republic. In 1511, a royal council gave him power over several islands. These included Hispaniola, Puerto Rico, Jamaica, and Cuba. However, the council said he did not own Uraba or Veragua.

Life in the islands was often difficult for Diego. He had many arguments with royal officials. These fights led King Ferdinand to recall him to Spain in 1514. Diego spent five years in Spain trying to fix his claims. King Charles finally gave his powers back in 1520.

Diego returned to Santo Domingo in November of that year. He arrived during a time of revolt by native people.

There were also very hard times involving enslaved people. In December 1522, enslaved Jolof workers revolted on his sugar plantation. This was the first major slave rebellion in the Americas. Some people escaped into the mountains to start new communities. Diego also faced trouble with a failed settlement in the Cumana area. These problems caused the king to lose trust in him. Diego died in Spain on February 23, 1526. His family continued to argue over his titles for a long time.
