Throughout her expansive career, she has appeared on more than 500 magazine covers. She achieved an unprecedented milestone by appearing on three consecutive Sports Illustrated Swimsuit Issue covers in 1979, 1980, and 1981. This feat was the first time any model had accomplished such a streak. Brinkley also secured a record-breaking 25-year contract with the cosmetics brand CoverGirl. In 2005, the company re-signed her to promote products for mature skin through magazine ads and television commercials. Her influence in the fashion industry is widely recognized by major global brands.

Brinkley's professional journey began through a chance encounter in a Parisian post office. In 1973, American photographer Errol Sawyer discovered her while she was studying art in Paris. Sawyer took her initial modeling photographs and introduced her to John Casablancas of the Elite Model Management agency. After meeting with fashion photographers Patrick Demarchelier and Mike Reinhardt, Brinkley returned to California. She met with Nina Blanchard, an Eileen Ford affiliate in Los Angeles. By the end of a single lunch meeting, she was booked for three national advertising campaigns. This rapid progression transformed her from a California surfer girl into a professional model.

Beyond the camera, Brinkley has built a diverse career in acting and stage performance. She made her film debut in the 1983 movie National Lampoon's Vacation as "the girl in the red Ferrari."
