Politkovskaya's career involved many different stages and roles. She was born in New York City in 1958 to Ukrainian Soviet diplomats. She grew up mostly in Moscow and studied journalism at Moscow State University. She graduated in 1980 with a thesis on the poetry of Marina Tsvetaeva. Before becoming a major reporter, she held various positions. She worked at the newspaper Izvestia and later at an aviation magazine. This magazine job provided her with unlimited air tickets. These tickets allowed her to travel widely across Russia to observe society.

Her most significant work began in the mid-1990s. She worked as an assistant editor for the newspaper Eskart. Later, she joined the newspaper Novaya Gazeta. This publication was known for critical investigative coverage of Russian politics. From 1999 to 2006, she wrote columns that challenged the post-Soviet regime. She also published several books based on her investigations. These included "A Dirty War" and "A Small Corner of Hell." In 2004, she published "Putin's Russia" for Western readers. This book provided her personal account of life in a changing nation.

Politkovskaya's reporting mechanism involved direct, hands-on investigation. She did not just observe from a distance. She traveled to Chechnya and the neighboring region of Ingushetia. She visited hospitals and refugee camps to interview those injured by fighting. She spoke with people who had been uprooted from their homes. In 1999, she even helped evacuate elderly residents from a home in Grozny. Her writing described the actions of Russian military forces and Chechen rebels. She also investigated the local administration led by Akhmad Kadyrov and his son, Ramzan Kadyrov.

Russian military forces once arrested her and subjected her to a mock execution. In 2004, she was poisoned while flying to help resolve the Beslan school hostage crisis. This required intensive medical treatment in Moscow to restore her health. Despite these threats, she refused to stop her work. She believed that society must not show apathy toward the loss of civil liberties. She often warned that a lack of information would lead to a "Soviet abyss."

On October 7, 2006, Anna Politkovskaya was murdered. She was found in the elevator of her apartment block in Moscow. This event happened on the 54th birthday of President Vladimir Putin. Her assassination drew massive international attention. In 2008, the Office of the Prosecutor General finished an investigation into the killing. Three men were accused of involvement, but they denied the charges. In 2014, five men were sentenced to prison for the murder. However, it remains unclear who ordered or paid for the killing.
