His naval career involved complex strategies and new technologies. Doria helped develop amphibious warfare for Spanish and Italian forces. This allowed his ships to assault coastal fortifications more directly.
Doria's early life was marked by service to many different masters. He was born in Oneglia to the noble Doria di Oneglia family. After being orphaned, he became a soldier of fortune. He served in the papal guard and for various Italian princes. In 1503, he fought in Corsica for the Genoese Navy. At age 46, he was appointed admiral of Genoa. Despite having no prior naval experience, he reorganized the entire Genoese fleet. He fought against the Ottoman Empire and Barbary pirates to protect his interests.

Doria used his connection to the Emperor to protect Genoa's independence. He also exerted immense influence over the republic's councils. He did not seek the title of Doge, the city's leader. These clans included families like the Cybo, Fieschi, and Spinola. He also reformed the office of the Doge. He reduced the term of office to two years instead of for life. He also made plebeians, or common citizens, ineligible for the position. These political structures lasted until the republic ended in 1797.
His military successes were often quite large in scale. In 1532, he led a fleet of 48 galleys to attack Ottoman positions. This helped prevent a potential conquest of Hungary.
