He grew up in Louisville, Kentucky. He was the sixth child in a large family. Flexner started his own private school for children. His school was different from others. It did not use grades or tests. Instead, students worked in small groups. They learned by doing hands-on work. Many students from his school went to top colleges.

Later, Flexner looked at how doctors were trained. In 1910, he published the Flexner Report. This report changed medical schools in the US and Canada. It made doctor training much harder and better. However, his ideas were also unfair. His report led to the closing of many schools for Black students.

Flexner also helped start the Institute for Advanced Study. This was a place for great thinkers to work together.
