The history of the university began with several long steps. In 1398, King Martí of Aragon wrote a letter to the councillors of Barcelona. He stated that he sought permission from the Pope to found a university. On 10 January 1401, he founded the Estudi General of Medicine. He gave this school the same privileges as the University of Montpellier. In 1450, King Alfons el Magnànim officially granted the prerogative to found a university. This happened because the Consell de Cent, or Barcelona's Council of One Hundred, requested it. This official foundation in 1450 was the culmination of a process started in 1398.

However, the university faced many periods of struggle and closure. During the reign of John II, a civil war caused delays in development. In 1714, the university was closed by the Bourbon dynasty following the War of the Spanish Succession. The authorities ordered the transfer of several faculties to the city of Cervera as punishment. The university remained closed until the liberal revolution during the reign of Isabella II. In 1837, the University of Cervera was transferred back to Barcelona. This event established the university as the cultural home for Catalonia and the Balearic Islands.

As the student population grew, the university had to build new homes. In 1863, work began on a large historic building designed by Elies Rogent. This construction took twenty years and was not finished until 1882. This project was important because it was one of the first buildings built outside the ancient city walls. To handle even more growth, the university started the Pedralbes district in 1952. The Faculty of Pharmacy was the first building completed there in 1956. Other buildings, like the Faculty of Law and the Faculty of Economics, followed in the coming years.
