Total synthesis

history Maturity 11-13 Next article →

Total synthesis is a special kind of chemistry. It is the art of building complex molecules from scratch. Scientists often try to make things found in nature. These natural products can come from plants, fungi, or the sea. They can even be tiny parts of animals. Sometimes, scientists make these things in a lab using simple starting materials. This is different from semisynthesis. In semisynthesis, they only build part of the molecule. Total synthesis aims to build the whole thing from the very beginning.

Building these molecules is like solving a huge puzzle. Scientists use a method called retrosynthetic analysis to start. They look at the final goal and work backward. This helps them plan the best path to build it. They must also use stereochemical control. This means they put the atoms in the exact right three-dimensional spots. If the shape is wrong, the molecule might not work. They also work to optimize reactions. This makes the process faster and helps them make more of the product.

This field has a very long and interesting history. In 1828, Friedrich Wöhler made urea from simple inorganic parts. This was a huge milestone for chemistry. Later, chemists worked to make camphor, which is a rare natural product. In 1904, Gustav Komppa helped find its structure. By 1907, he could make it in a factory in Finland. In 1954, Vincent du Vigneaud made hormone molecules called oxytocin and vasopressin. He even won a Nobel Prize for this work in 1955.

Many famous scientists have led this work. Robert Burns Woodward was a very important leader in this field. He made many things like cholesterol in 1951 and quinine in 1944. He also made chlorophyll in 1960. In 1972, his team finished the synthesis of Vitamin B12. This was a giant task with over 100 steps. It involved more than 100 researchers from 19 different countries. Another great chemist, Elias James Corey, won a Nobel Prize in 1990 for his work on planning these paths.

Today, total synthesis helps us in many ways. It is a key tool for making new medicines. For example, scientists made the anticancer drug Taxol in the 1990s. This work also helps us understand how living things work. It helps researchers study biological systems and processes. Modern scientists now use new tools like biocatalysis. This combines old methods with biology to make things better. The field continues to grow and help us understand the diversity of nature.
