The history of San Luis began in the late 1500s. Luis Jufré de Loaysa y Meneses founded the city of San Luis in 1594. However, the settlement was abandoned shortly after its founding. A man named Martín García Óñez de Loyola refounded the area in 1596. He gave the new settlement the name San Luis de Loyola.

Since Argentina returned to democratic rule in 1983, the Rodríguez Saá family has been very influential. This family belongs to the Peronist political group. They have held the position of governor many times. Adolfo Rodríguez Saá was one such former governor. He also served as a Senator.
