She was born in Frankfurt. Later, she moved to Vienna to study her voice. She also studied in Paris.

Mathilde taught many great singers. Her most famous student was Nellie Melba. She also taught Emma Calvé and Frances Alda. Her daughter, Blanche Marchesi, was also a noted singer.

Mathilde had special ways to teach. She used an analytical method. This meant students had to understand the music they sang. She did not want them to just repeat notes. She also liked a slow way of learning. She told new students to practice for only five minutes at a time. They would do this three or four times each day. As they got better, they could practice for longer.

Mathilde loved the bel canto style. This is a beautiful way of singing. She believed singers should breathe in a natural way. She also cared a lot about vocal registration. This is how the voice develops. Her hard work helped many people become stars.
