According to the 2021 Canadian census, the city had 422,324 residents. The larger metropolitan area reached a population of 543,551. This growth happened as the city annexed many surrounding communities.

The history of the region begins long before European arrival. Archaeological sites known as the Parkhill Complex show Paleo-Indians lived here 11,000 years ago.

Just before European settlement, the area held villages of the Attawandaron, Odawa, and Ojibwe. The Lawson Site contains a partial reconstruction of a Neutral Iroquoian village. This village was approximately 500 years old and held about 2,000 people. Later, the Iroquois drove these groups out during the Beaver Wars. The Iroquois eventually left the region after 50 years. They were then driven out by the Ojibwa. An Anishinaabeg community once lived near the river forks. They called the area Eshkani-ziibi, which means Antler River. They also used the name Baketigweyaang, meaning "At the River Fork."

European involvement began in the late 18th century. In 1793, Lieutenant-Governor John Graves Simcoe chose this site. He wanted it to be the capital of Upper Canada.

Simcoe named the city and river after places in England. He originally intended to call the site Georgiana. However, it became an administrative seat instead of a capital. The 1796 London Township Treaty with the Chippewa ceded the north bank of the Thames to Upper Canada. The area was part of the Talbot Settlement. Colonel Thomas Talbot oversaw land surveying and built the first government buildings. Peter Hagerman established the first European settlement between 1801 and 1804. The village was officially founded in 1826 and incorporated in 1855.

London faced many challenges during its early development. In 1832, an outbreak of cholera hit the new settlement. During the Upper Canada Rebellion of 1837, London showed strong Tory support. This led the British government to place a garrison there in 1838. This arrival increased the population with soldiers and their dependents. A massive fire occurred on 13 April 1845. Most buildings were made of wood at that time. The fire destroyed 150 buildings and nearly one fifth of the city.

This was the first million-dollar fire in the province. It even destroyed the town's only fire engine. Despite this, most buildings were rebuilt by 1846.

As the city grew, it absorbed several smaller municipalities. The village of Petersville was founded by Samuel Peters in 1853.

Kensington was founded around 1878. These two areas amalgamated in 1881 to form London West. This area faced repeated flooding, including a serious event in July 1883. London West was eventually annexed to London in 1897. Another area, Lilley's Corners, was founded by Charles Lilley in 1854. It became a village in 1872 and was annexed in 1885. The southern area, known as London South, was annexed in 1890. This included Wortley Village, which still maintains a distinct sense of place.
