Early on, he worked with famous Pakistani lawyers like Huseyn Shaheed Suhrawardy. He often handled cases involving missing persons during the military rule of Ayub Khan. Between 1961 and 1968, he taught law at Dhaka University. Later, he served as the chief political negotiator for the Awami League. This role required him to lead discussions regarding the transfer of power in 1971. He was also a member of the National Assembly of Pakistan.

Throughout his life, Hossain has specialized in several distinct legal fields. He is a leading expert in human rights and international arbitration. International arbitration is a way to settle disputes between parties without a traditional court. He also works extensively in energy law and corporate law. His expertise in energy law led to the creation of the Bangladesh Petroleum Act. Additionally, he has worked on maritime disputes. These are legal disagreements regarding ocean boundaries between nations like Malaysia and Singapore. He has also served as a UN Special Rapporteur for Afghanistan.

The year 1971 was a major turning point in his life and for his country. During the Bangladesh War of Independence, Hossain was imprisoned in West Pakistan. He was released in late December 1971. Following the war, he helped form the first post-independence government. From 1972 to 1975, he served as the Law Minister. He also chaired the committee responsible for drafting the new constitution. At 35 years old, he was one of the youngest constitution drafters in the world. The resulting 1972 Constitution made Bangladesh the first constitutionally secular state in South Asia.

The impact of his legal work can be seen in other nations as well. His reforms in Bangladesh were emulated by both India and China. Specifically, India used his ideas in its 42nd constitutional amendment. China also used his models during its reform and opening up of energy law. While some of his reforms were repealed in 1977, secularism was later reinstated by the Supreme Court in 2010. His life has been described by journalists as the "conscience of the nation." He remains a significant figure in the history of South Asian law and politics.
