His transition to film occurred through small roles in movies like West Side Story in 1961. His ability to perform comedy was eventually noticed by actor Tony Randall. This led to guest appearances on various sitcoms. These included Dennis the Menace, The Donna Reed Show, and Harrigan and Son.

In the early 1960s, Astin achieved his first major television lead. He starred in the ABC sitcom I'm Dickens, He's Fenster during the 1962–63 season. He played the character Harry Dickens alongside Marty Ingels. The show combined witty dialogue with slapstick comedy. Although the show received critical praise, it faced tough competition. It aired against high-rated programs like Sing Along with Mitch and Route 66. Despite being canceled by ABC, the show eventually gained a following. A total of 32 episodes were produced for this series.

Beyond acting, Astin proved his skills as a director and writer. He made his directorial debut with a comedic short film titled Prelude in 1968. Education became a significant second chapter for Astin. He returned to his alma mater, Johns Hopkins University, to teach. This endowed chair aims to make theater a major program for undergraduates.
