Trypsin is a proteolysis enzyme that breaks down insulin before it can be collected. Banting realized he needed to stop the trypsin from working first. He read about a method to close the pancreatic duct using a ligature. This procedure would cause the trypsin-secreting cells to die off. Once those cells were gone, the insulin could be safely extracted from the islets of Langerhans.

Banting worked with John Macleod at the University of Toronto. He also worked with a student named Charles Best. To get enough material, they used the pancreases of fetal calves from a slaughterhouse. They found these extracts were just as potent as those from dogs. By December 1921, they had successfully extracted insulin from adult pancreases as well. On January 11, 1922, 14-year-old Leonard Thompson received the first insulin injection. This marked a massive turning point for medical science.

Banting's achievements earned him the highest honors in science. In 1923, he and John Macleod were awarded the Nobel Prize in Physiology or Medicine.
