The history of the area spans many millennia. Human settlements appeared during the Paleolithic era. Archeological finds show people lived on Monastyrskyi Island. During the Bronze Age, diverse tribes settled the land. The Scythian empire controlled the area from the 1st to the 3rd century BC. Later, the Kievan Rus' period saw fighting between various groups. These included the Khazars, Pechenegs, and Cumans. The Mongol Empire conquest in the 13th century devastated the region. Eventually, the area became part of the Polish-Lithuanian Commonwealth.

Cossack communities settled the site by at least 1524. They established a trading settlement at an important river crossing. In 1635, the Polish-Lithuanian Commonwealth built the Kodak Fortress. This was located above the Dnipro Rapids.

The fortress was destroyed by Cossacks but rebuilt in 1645. A settlement grew around it known as Novyi Kodak. This town had a population of at least 3,000 people. The Kodak Palanka seal later became part of the city's official coat of arms.

Modern Dnipro began with a formal imperial decree. In 1784, Empress Catherine the Great ordered a new city. She moved the provincial center to the right bank of the Dnipro. The city was named Yekaterinoslav to honor her.
