Ammonium acetate

physical science Maturity 11-13 Next article →

Ammonium acetate is a chemical compound with the formula NH4CH3CO2. It appears as a white, hygroscopic solid. Being hygroscopic means this substance naturally absorbs moisture from the surrounding air.

This property makes it quite difficult to obtain the substance in a crystalline form. It is a versatile tool used across many scientific and industrial fields. Scientists use it in laboratories, food production, and even in agriculture. Its unique chemical properties allow it to serve many different roles.

There are two primary ways to produce ammonium acetate through chemical reactions. One method involves the neutralization of acetic acid with ammonium carbonate. Another common way to create it is by saturating glacial acetic acid with ammonia. You can also derive the compound from a reaction between ammonia and acetic acid.

These reactions allow the compound to be made for commercial availability. Once created, the substance can be used in a wide variety of chemical processes.

A key feature of ammonium acetate is its ability to act as a buffer. A buffer is a substance that helps maintain a stable pH level in a solution. This compound is the salt of a weak base, ammonium, and a weak acid, acetic acid. When it is dissolved in pure water, the solution typically reaches a pH of 7. This happens because the acetate and ammonium parts neutralize each other.

However, it functions as a dual component buffer system. It buffers around pH 4.75 ± 1 for acetate and pH 9.25 ± 1 for ammonium. It actually has no significant buffer capacity at pH 7, which is a common misconception.

Because ammonium acetate is volatile at low pressures, it is very useful in specialized testing. Volatility means the substance can turn into a gas more easily under certain conditions.

This makes it an excellent replacement for cell buffers that contain non-volatile salts. Specifically, it is used when preparing samples for mass spectrometry. It is also a popular buffer for mobile phases in High-Performance Liquid Chromatography, or HPLC. It works well with detection methods known as ELSD and CAD-based detection. Other volatile salts, such as ammonium formate, have also been used for these tasks.

In the world of organic synthesis, ammonium acetate serves several specific roles. It acts as the main precursor to acetamide, which has the formula CH3C(O)NH2. The process involves the reaction NH4CH3CO2 → CH3C(O)NH2 + H2O. It also functions as a catalyst in a process called the Knoevenagel condensation. Furthermore, it serves as a source of ammonia in the Borch reaction.

These reactions are essential for building complex organic molecules in a lab.

Ammonium acetate has many practical applications outside of pure chemistry labs. In agriculture, it is used to determine the cation exchange capacity, or CEC, of soil. It is also used to find the amount of available potassium in soil. In this process, the ammonium ion acts as a replacement cation for potassium. The compound also serves as a biodegradable de-icing agent.

Additionally, it is used as a protein precipitating reagent during dialysis to remove contaminants via diffusion.

History provides an interesting name for this compound. It is sometimes referred to as the "spirit of Mindererus" in aqueous solution. This name honors R. Minderer, who was a physician from Augsburg.

Beyond the lab, the compound is even found in the food industry. It is used as an acidity regulator and is assigned the INS number 264. This food additive is approved for use in both Australia and New Zealand. It even plays a role in art conservation through Calley's method for preserving lead artifacts.
