N'Dour's musical career began in his youth in Dakar. He started performing music when he was only 12 years old. At age 15, he joined a band called Diamono. In 1975, he toured West Africa with this group. By 1976, at age 16, he signed with the Star Band. He performed at the Miami club in Dakar. This period helped him become a sensation. In 1978, he joined musicians to form Étoile de Dakar. This band was vital to the evolution of mbalax. Eventually, the group split due to internal problems. N'Dour then formed Super Étoile de Dakar. This became his long-term backing band.

His cultural identity is a blend of different traditions. N'Dour was born to a Serer father and a Toucouleur mother. In the Senegambia region, ethnicity is often determined by the father's line. Therefore, N'Dour is ethnically Serer. However, he is culturally Wolof. His parents encouraged a modern outlook. This allowed him to act as a "modern griot." A griot is a traditional storyteller or musician. N'Dour is also a Mouride disciple. This means he belongs to the Mouride brotherhood. This is one of the four large Sufi orders in Senegambia. Because of this, he often uses Islamic chants in his music.

N'Dour has achieved massive international success. In 1994, he released the trilingual single "7 Seconds." This duet with Neneh Cherry became his biggest international hit. He also wrote the official anthem for the 1998 FIFA World Cup. His voice has been described as an "arresting tenor." In 2005, he won a Grammy Award for his album Egypt. This was his first American Grammy. In 2013, he shared the Polar Music Prize. This prize was worth $150,000. It honored his work in promoting understanding between faiths.
