Sykes utilizes several different methods to perform her craft. She performs stand-up comedy, which involves telling jokes to a live audience. She also works as a television actress, playing specific characters in scripted series. In addition to live action, she is a voice actress for animated films. This role requires using her voice to provide personality to drawn characters. She has also worked as a comedy writer and a late-night talk show host. Her career demonstrates how one person can master many different forms of comedy.

Her professional work can be divided into several distinct areas. In television, she has held recurring roles in shows like Curb Your Enthusiasm and The New Adventures of Old Christine. Sykes' journey to fame began with a very different professional path. She was born in Portsmouth, Virginia, on March 7, 1964. Her family moved to Maryland during her third-grade year. After graduating from Hampton University with a degree in marketing, she began a stable career. She worked as a contracting specialist at the National Security Agency for five years. Her family history is a subject of significant historical interest. Her ancestry was featured on the PBS program Finding Your Roots with Henry Louis Gates Jr. Historian Ira Berlin noted that her family history is unique in American records. He stated it is the only case he knows where a Black family's roots in freedom can be traced back to the late 17th century. This history traces back to a 1683 court case involving her ancestor, Elizabeth Banks. Elizabeth was a free white woman and an indentured servant. Her daughter, Mary Banks, inherited her mother's free status.
