He is most recognized for his extensive work in English dubbing. This process involves recording English dialogue for Japanese anime series and video games. Mignogna has contributed his voice to over 100 different titles. His performances include major series such as Dragon Ball Z and Bleach. He earned the American Anime Award for Best Actor in 2007. This prestigious award recognized his performance as Edward Elric in the Fullmetal Alchemist franchise.

Voice acting requires immense skill to bring animated characters to life. Mignogna has demonstrated this versatility across many different genres. He voiced the powerful Broly in the Dragon Ball films and Dragon Ball Super: Broly. He also portrayed Tamaki Suoh in the series Ouran High School Host Club. In the realm of video games, he provided the voice for E-123 Omega in the Sonic the Hedgehog series. He also voiced Junpei Iori in the game Persona 3. His ability to shift tones allows him to play characters ranging from heroes to villains.

Mignogna's career path was shaped by his early interests in media. He grew up watching classic animated shows like Speed Racer. He was also a dedicated fan of the science fiction series Star Trek: The Original Series. He pursued formal education at Liberty University, earning a degree in television and film production. Before entering voice acting, he taught English and speech at Trinity Christian Academy in Florida. In 1990, he moved to Houston, Texas, to teach film and video production at The Art Institute of Houston.

His entry into the anime industry began through his work in video production. While working with John Gremillion in Houston, he became involved with ADV Films. He made his debut as the character Vega in the Street Fighter II V series. Mignogna eventually became a frequent presence at anime conventions. At one time, he attended between 15 and 25 conventions every year. He set a professional goal to record at every major dubbing location. He was the first ADV voice actor to record at Funimation in Dallas and later in New York.

Beyond voice acting, Mignogna has maintained a significant career in music. He has released eight studio albums and eight audio recordings. His musical background includes producing hundreds of jingles for various commercials. In 1993, he appeared as a contestant on the television talent show Star Search. During that appearance, he performed the song "Worth Waiting For." He has also performed the U.S. national anthem at several Houston Astros baseball games. His musical work even extends to anime, including English cover versions of songs from One Piece.

In addition to animation, Mignogna has worked in fan-led live-action productions. He participated in several Star Trek fan projects, including Star Trek Continues. In that series, he directed and starred as Captain James T. Kirk. Critics gave him positive reviews for his portrayal of the character. He used the unique mannerisms and cadence of William Shatner to inform his performance. He also held roles such as an Andorian Captain and a Klingon named Malkthon in other productions. These projects show his interest in directing and executive producing.

In early 2019, Mignogna's career faced significant challenges due to serious allegations. During the #MeToo movement, several fans and fellow voice actors accused him of sexual harassment. Mignogna denied all accusations of misconduct. Following an internal investigation by Sony Pictures, Funimation dismissed him from their productions. Rooster Teeth also cut ties with him in February 2019. Mignogna pursued legal action for defamation against Funimation and his accusers. However, the courts eventually dismissed his claims under free speech protections.
