During his first audition, Tilde De Rensis played the piano. They married in 1937 and became partners in life.

Gobbi's professional career began in the 1930s. He made his debut in Gubbio in 1935. He sang the role of Count Rodolfo in La sonnambula. Later, he worked at La Scala in Milan. He served as an understudy during the 1935–1936 season. This helped him gain a lot of experience. By 1937, he sang in Rome at Teatro Adriano. He also gained experience at La Scala in Milan as an understudy. In 1937, he sang the role of Germont in La traviata in Rome. Under the guidance of conductor Tullio Serafin, Gobbi prepared many of his most important roles. These included Scarpia, Rigoletto, and Simon Boccanegra. He performed in many major Italian venues, such as La Fenice in Venice and the Teatro Communale in Florence.

After World War II, Gobbi's career became truly international. He appeared at the San Francisco Opera in 1948. He first performed at London's Royal Opera House, Covent Garden, in 1950. He had a long relationship with the Lyric Opera of Chicago, performing there from 1954 to 1974. In 1956, he made his debut at the Metropolitan Opera in New York. He also appeared in over 25 films. Sometimes he acted in motion pictures, and other times he provided the singing voice for actors. For example, he provided the singing voice for Anthony Quinn in the 1953 film Cavalleria rusticana.

One of Gobbi's most famous roles was Scarpia in Puccini's Tosca. He claimed to have sung this specific part nearly 1,000 times. He worked closely with the famous soprano Maria Callas. They recorded a classic version of Tosca in Milan in 1953. This recording featured Giuseppe Di Stefano and was conducted by Giuseppe Di Stefano. Many people consider this 1953 mono recording to be one of the finest complete opera recordings ever made. Gobbi was a close friend and admirer of Callas. He once said that performing with her was not just performing, but "living."

In the 1960s, Gobbi expanded his work into stage directing. He directed about ten different operas between 1965 and 1982. These productions were staged nearly 40 times across Europe and North America. He directed many productions for the Lyric Opera of Chicago, including nine of his ten staged operas. His directed works included titles like Otello and Gianni Schicchi. He even sang the title role in three of the productions he directed. This showed his deep love for opera both on the stage and on recorded discs.

Gobbi left behind a rich legacy through books and family. In retirement, he wrote his autobiography, titled Tito Gobbi: My Life, with the help of Ida Cook. He also wrote Tito Gobbi and His World of Italian Opera. This second book examined the operas he performed or directed. His daughter, Cecilia, continues his work today. She runs the "Associazione Musicale Tito Gobbi." This organization is dedicated to preserving and celebrating his massive contribution to the world of opera.
