Her musical training involved several different teachers and locations. She began her studies with Josef Neruda, a cellist in her hometown. Later, she attended the Prague Conservatory to study under Giovanni Battista Gardigiani. After being expelled from the conservatory in 1851, she studied with Vojtěch Čaboun. She eventually moved to Trieste to live with her brother. There, she studied with Luigi Ricci, a conductor who had worked with Verdi. Finally, she moved to Milan to study with Francesco Lamperti.

Stolz's career spanned many different types of operatic roles. She performed title roles in famous works like Donizetti's Lucrezia Borgia and Bellini's Norma. She also sang Mathilde in Rossini's Guillaume Tell and Alice in Meyerbeer's Robert le diable. Within Verdi's own compositions, she sang roles such as Amelia in Un ballo in maschera and Gilda in Rigoletto. These roles required a wide range of emotional and technical abilities. Each performance helped establish her reputation across Europe.

Her professional history is marked by several historic musical premieres. In 1865, she made her debut at the famous La Scala opera house in Milan. She sang the title role in Verdi's Giovanna d'Arco during that appearance. In 1867, she performed as Elisabetta in the first Italian performance of Don Carlo in Bologna. On February 27, 1869, she sang Leonora in a revised version of La forza del destino in Milan. These events placed her at the center of the most important musical developments of her time.

One of her most significant achievements involved the opera Aida. On February 8, 1872, Stolz was the first person to sing the title role of Aida in Italy. This performance took place at La Scala and served as the European premiere. Although the opera had premiered in Cairo the previous December, Verdi considered the Milan performance to be its real premiere. He was heavily involved in every stage of this production. Stolz continued to perform this role in Vienna in 1875 and in Paris in 1876.

Stolz also played a vital role in the performance of Verdi's Requiem. This is a large-scale musical composition written for voices and instruments. She was the soprano soloist at the work's premiere on May 22, 1874. In 1875, she performed the Requiem at the Royal Albert Hall in London. During that concert, Verdi himself served as the conductor. Her ability to handle such complex music proved her status as a leading singer.

Her personal life was also closely linked to the world of music and Verdi. She was once the fiancée of the conductor Angelo Mariani. After Mariani, she became the companion of Giuseppe Verdi following the death of his wife in 1897. She lived with him at his home in Sant'Agata. Stolz died in Milan in 1902, just one year after Verdi passed away. Her legacy is honored by a hall named after her in Kostelec nad Labem.

Her family was also deeply involved in the musical profession. Stolz had two older twin sisters, Francesca and Ludmila, who both became sopranos. Her nephew, Luigi Ricci, became a conductor and a composer. He eventually inherited her estate and changed his name to Luigi Ricci-Stolz. This connection shows how music was a central part of her entire family lineage.
