It landed three weeks before its twin, Opportunity. The landing site was named Columbia Memorial Station. This name honored seven astronauts from the Space Shuttle Columbia.

Spirit was one of two rovers sent to study the red planet. Its twin, Opportunity, landed on the other side of Mars three weeks later. The name "Spirit" was chosen through a student essay competition sponsored by NASA. This rover played a vital role in helping scientists understand the history of the Martian environment.

The primary scientific goal was to search for clues regarding past water activity on Mars. Scientists wanted to find rocks and soils that held evidence of water. They specifically looked for minerals deposited by processes like precipitation or evaporation. They also sought iron-bearing carbonates, which are minerals that form in water. By studying these materials, researchers could determine if ancient Martian environments were suitable for life. Additionally, Spirit helped calibrate instruments on the Mars Reconnaissance Orbiter (MRO). This ensured that observations made from orbit were accurate and effective.

Spirit was much more successful than NASA originally anticipated. The mission was initially planned to last only 90 sols, which is about 92.5 Earth days.

However, due to cleaning events that cleared dust from its solar panels, Spirit functioned for 2,208 sols. This means it operated for over twenty times its expected lifespan. By August 2007, it had set a record for the longest operational duration for a lander or rover on Mars at that time. It drove much further than the planned distance, allowing for much deeper geological analysis of the planet.

Challenges arose in May 2009 when Spirit became stuck in soft sand at a location called Troy.

The rover was resting on a layer of iron(III) sulfate, also known as jarosite, which was hidden under the soil. This substance has very little cohesion, meaning it does not stick together well. Because the sand was so loose, the wheels could not gain traction. NASA spent eight months attempting to program extrication drives to free the rover. By January 26, 2010, officials realized the rover was likely irrecoverably obstructed. Spirit continued to work as a stationary science platform for a short time afterward.

The mission eventually came to an end due to the extreme Martian environment. The last communication from Spirit occurred on March 22, 2010. Scientists believe the rover entered a low-power hibernation mode or suffered a mission clock fault. Because it was stuck on flat ground rather than a sun-facing slope, it could not keep its internal temperature stable. It is estimated that its temperature dropped to levels that damaged critical components. NASA continued trying to regain contact until May 24, 2011. On that day, the mission was officially declared complete, marking the end of a historic era of Martian exploration.
