Mitochondrial DNA research shows that Aboriginal people reached the Eyre Peninsula between 45,000 and 49,000 years ago. They arrived from both the east and the west. On the island of Kangaroo Island, people lived long before rising sea levels cut the island off from the mainland. European contact began much later in the 17th century. In 1627, a Dutch ship called the Gulden Zeepaert mapped part of the coastline.

The formal establishment of South Australia followed a very different path than other Australian colonies. In 1788, the land was claimed by Britain as part of New South Wales. However, the British Parliament passed the South Australia Act 1834 to create a new province. This act allowed the state to be a "convict-free" colony. This means it was intended for free immigrants rather than prisoners. The settlement followed the theory of "systematic colonisation" proposed by Edward Gibbon Wakefield. The goal was to create a center of civilization with religious tolerance and civil liberties.

Colonial government officially began on 28 December 1836, a date known as Proclamation Day. Before Adelaide was built, the South Australian Company established a temporary settlement at Kingscote on Kangaroo Island. Colonel William Light led the first expedition to survey the coast. He was tasked with finding the best site for the capital city. Light planned the city of Adelaide using one-acre town sections and 134-acre country sections. While the province was meant to protect Aboriginal land rights through the Letters Patent, these rights were often ignored by squatters. This led to many conflicts during the Australian Frontier Wars.

Today, the population of South Australia is approximately 1.9 million people as of June 2025. The state is highly centralized, meaning most people live in one place. More than 67% of the population resides in the metropolitan area of Adelaide.

The second-largest population center is Mount Gambier, which has 26,878 residents. Most other people live in fertile areas along the River Murray or the southeastern coast.

The economy is driven by three main industries: agriculture, manufacturing, and mining. The healthcare sector is also a major employer for the people living there.

South Australia is internationally famous for its agricultural production, especially its wine. It produces more than half of all the wine in Australia.

Major wine-growing regions include the Barossa Valley, Clare Valley, and the McLaren Vale.
