The history of Santa Clara began on 15 July 1689. A group of 175 people founded the settlement. This group included 138 people from two large families who lived on nearby land. The remaining 37 people came from seven other families in the coastal city of San Juan de los Remedios. These families chose to move to the interior to avoid being besieged by pirates. They arrived at a hill on 1 June 1689. Tradition says they celebrated a mass under a tamarind tree to found the city.

This site is known as Loma del Carmen. The city went through many name changes, such as Cayo Nuevo and Dos Cayos, before becoming Santa Clara.

The city was designed using a specific Spanish standard. This involved a perfect squared layout with a central plaza. This plaza is known today as Parque Vidal. The first buildings built were the cabildo, which is a city council, and a church made of palm trees. In 1725, the palm church was replaced by a brick building called the Catedral de Santa Clara de Asís. This cathedral stood in the center of Parque Vidal until 22 August 1923. It was demolished by a mayor to expand the plaza. This decision remains a topic of criticism today because the church was a major colonial structure.

Santa Clara is closely linked to two famous historical figures. Marta Abreu de Estévez is known as the "Benefactress of the City." She and her husband, Luis Estévez, supported Cuban rebels during the War of Cuban Independence. Abreu used her own wealth for philanthropy to improve the city. She funded an electric plant, schools, an asylum, and a fire station. She also built the Teatro La Caridad, which is one of seven major colonial-era theaters still standing in Cuba. She even donated her palace, which now serves as the Biblioteca Martí.

The second famous figure is Ernesto Che Guevara, a leader of the Cuban Revolution. In late 1958, Guevara led a column of guerrillas in the Battle of Santa Clara. His forces used a bulldozer to destroy railroad tracks. This caused a train full of troops and supplies to derail. On 31 December 1958, Guevara and Camilo Cienfuegos attacked the city. The battle was chaotic and led to the capture of the city. This victory was a decisive moment that caused the Batista government to fall.

Because of its central position, Santa Clara is a vital transport hub. It is crossed by the Carretera Central highway and the Autopista A1 motorway. The Santa Clara railway station is part of a principal national line connecting Havana to Santiago. The city is also served by the Abel Santamaría Airport. The city is divided into several administrative areas called repartos. These include urban areas like Centro and Vigía, semi-urban areas like Universidad, and rural areas like San Miguel.

Education is a major part of the city's identity through the University "Marta Abreu" of Las Villas. The university offers many specialized faculties.
