Most of the urban area lies within the Plasy Uplands. However, some parts of the municipal territory reach into the Švihov Highlands to the south and east. The highest elevation in the city is the hill known as Chlum. The lowest point is found in the river bed of the Berounka.

The history of Plzeň is marked by its role as a strategic center for trade and defense. While a castle was mentioned as early as 976, the city of New Plzeň was officially founded in 1295 by King Wenceslaus II.

This new settlement, which became known as Plzeň, sat on important trade routes connecting Bohemia with Bavaria. By the 14th century, it had grown into the third-largest city in Bohemia. During the 15th-century Hussite Wars, the city became a famous center of Catholic resistance. It survived three separate sieges led by Prokop the Great. Later, during the 17th-century Thirty Years' War, the city faced occupation. It was captured by Mansfeld in 1618 and was not regained by Imperial troops until 1621. The city's architecture began to reflect the Baroque style toward the end of the 17th century.

In the 19th century, Plzeň underwent a period of rapid industrialization. This shift changed the city from a trade hub into an engineering powerhouse. In 1869, Emil Škoda founded the Škoda Works.

This company became one of the most influential engineering firms in Austria-Hungary and later in Czechoslovakia. By 1917, the Škoda Works employed more than 30,000 workers. The city also became a massive hub for rail transport. The National Railways train workshop was the largest rail repair shop in all of Austria-Hungary. The completion of the Plzeň railway junction between 1861 and 1877 helped drive this growth. This industrial boom also led to the expansion of the local Czech population and the rise of the urban poor.

The city also faced significant challenges during the era of World War II. Following the Munich Agreement in 1938, Plzeň became a frontier town as Nazi Germany's borders moved closer. During the German occupation from 1939 to 1945, the Škoda Works was forced to produce armaments for the Wehrmacht. The Nazi regime also operated a Gestapo prison and a forced labour camp in the Karlov district. In January 1942, over 2,000 Jewish residents were deported to the Theresienstadt concentration camp. As the war ended, the city suffered devastating air raids. British Royal Air Force bombers targeted the marshalling yard in April 1945. Shortly after, the U.S. Eighth Air Force conducted its last heavy bomber mission in Europe against the Škoda Works. On 6 May 1945, the city was finally liberated by the 16th Armored Division of General George Patton's Third Army.

The city is organized into ten self-governing boroughs. These are further divided into 25 municipal parts. For example, the Plzeň 3 borough is the most populous, with 55,363 residents according to the 2021 census. Other notable areas include Bolevec, which has 29,697 inhabitants, and Severní Předměstí, with 18,875. The city's infrastructure supports a modern lifestyle, including a tram system that has operated since 1899.
